Challenge:<\/strong>\u00a0Type 2 diabetes (T2D) is the leading cause of cardiovascular and\u00a0renal\u00a0diseases. Although many treatments are available, they\u00a0only partially reduce\u00a0the risk of serious complications\u00a0in\u00a0diabetic\u00a0patients. As a first step towards reducing diabetes-related morbidity and mortality, the team had\u00a0developed a Polygenic Risk Score (PRS)\u00a0in\u00a0T2D Caucasian\u00a0patients\u00a0that combines\u00a0data on\u00a0sex, age of onset and diabetes duration to better predict risks of complications. Because ethnicity\u00a0and genetic variation play an important role in\u00a0the\u00a0development\u00a0of T2D\u00a0and\u00a0its\u00a0complications, the test would benefit from being personalized\u00a0according to\u00a0ethnic backgrounds.<\/p>\n\n\n\n

Solution:<\/strong> The\u00a0team extended\u00a0their\u00a0genomic\u00a0predictive tests\u00a0to major ethnic groups.\u00a0At the end of the project, beside\u00a0Caucasian\u00a0(Celtic\u00a0&\u00a0Slavic)\u00a0patients, the team had thus successfully analyzed\u00a0large\u00a0data of South European, South Asian and Afro-American\u00a0patients, while\u00a0data of\u00a0Middle East\u00a0and\u00a0Chinese patients are now being analyzed through new partnerships. Data\u00a0include\u00a0geo-ethnic background,\u00a0but also\u00a0sex, age of onset and diabetes duration. The resulting\u00a0predictive\u00a0test\u00a0is a meta-PRS, which combines\u00a0data from 10 PRSs, and was validated on large patient cohorts.\u00a0The team also\u00a0developed a\u00a0clinically actionable report\u00a0intended to health care providers.<\/p>\n\n\n\n

Achievements\/Impact:<\/strong>\u00a0The\u00a0project\u00a0developed\u00a0tools\u00a0to predict susceptibility to T2D\u00a0and its\u00a0vascular and renal\u00a0complications,\u00a0as well as\u00a0individual therapeutic responsiveness.\u00a0\u00a0A major accomplishment of the project is that OPTITHERA now offers direct-to-consumer (DTC) genetic health risk tests for patients diagnosed with T2D, for most ethnic groups, with an actionable report for their physicians. This precision medicine enables clinical practice to personalize health care for T2D patients and ultimately to prevent complications. A major economic impact is the reduction of the important costs associated to these diabetes complications.<\/p>\n\n\n\n
